Calibration of the XY Scanners

Calibration:

Start ACQ4, Move stage to get objective/position read.

• Set Camera to “CLEAR_MODE” = clear pre-exposure
• Start camera set binning to 4 (high frame rate).
• Make sure laser is visible, in focus and not clipping the camera. (you will need a fluorescent target for this)
• Click “calibrate” in the acqmanager

Do for both 5x and 40x objective (water immersion…). Then check the spot size.
